BriefDefinitive Report A -y/b T Cell Receptor Heterodimer Induces the Expression of CD4 and CD8 in Thymocytes

چکیده
CD4 and CD8 have been useful surface markers for a/R T cell maturation . In an a/(3 T cell receptor (TCR) transgenic SCID mice system, it has been shown that a/0 TCR alone is sufficient to induce CD4 and CD8 surface expression on thymic T cells. Although the late embryonic thymic y/b T cells are predominately single and double positive, it has not been clear if y/b TCR has a similar capacity. In this study, we show that when transgenes encoding the earliest embryonic y/b TCR are coexpressed with the SCID defect, the y/b transgenes promote the appearance of both the CD4 8 and CD4+8+ T cells in the thymus . Furthermore, the expression of CD4 and CD8 does not require continuous surface y/b TCR expression . These results indicate that y/bTCR alone can promote the CD4/8 surface expression, and may suggest a role for y/b T cells in initiating normal thymic ontogeny.

Comparative Analysis of CD4+ and CD8+ T Cells in Tumor Tissues, Lymph Nodes and the Peripheral Blood from Patients with Breast Cancer
Background: CD4+ and CD8+ T cells are the main types of lymphocytes in cell-mediated immunity and play a central role in the induction of efficient immune responses against tumors. The frequencies of T cell subtypes in the peripheral blood and tumor tissues, and draining lymph nodes (dLN) can be considered as useful markers for evaluation of the immune system in cancers. Methods: In this study,...

CD4 and CD8 have been useful surface markers for alpha/beta T cell maturation. In an alpha/beta T cell receptor (TCR) transgenic SCID mice system, it has been shown that alpha/beta TCR alone is sufficient to induce CD4 and CD8 surface expression on thymic T cells. Although the late embryonic thymic gamma/delta T cells are predominately single and double positive, it has not been clear if gamma/...
متن کاملEarly Deletion and Late Positive Selection of T-Cells Expressing a Male-Specific Receptor in T-Cell Receptor Transgenic Mice
The ontogeny of T cells in T-cell receptor (TCR) transgenic mice, which express a transgenic alpha beta heterodimer, specific for the male (H-Y) antigen in association with H-2Db, was determined. The transgenic alpha chain was expressed on about 10% of the fetal thymocytes on day 14 of gestation. About 50% of day-15 fetal thymocytes expressed both alpha and beta transchains and virtually all fe...
متن کاملRegulation of pre-T cell receptor (pT alpha-TCR beta) gene expression during human thymic development
In murine T cell development, early thymocytes that productively rearrange the T cell receptor (TCR) beta locus are selected to continue maturation, before TCR alpha expression, by means of a pre-TCR alpha- (pT alpha-) TCR beta heterodimer (pre-TCR). The aim of this study was to identify equivalent stages in human thymocyte development. We show here that variable-diversity-joining region TCR be...

Behcet disease (BD) is a systemic inflammatory disease of the unknown etiology. There is however, some evidence to suggest that immunological abnormalities are important in its pathogenesis, furthermore several T-cell abnormalities which may be quite relevant to autoimmune origin of the disease have been described. We report here our study of T-cell subsets, B and NK cells in 68 patients with B...
